Natural Culture Media
The most commonly used natural culture medium is serum, with calf serum being the most widely utilized. Serum contains multiple cell growth factors, adhesion-promoting factors, and various bioactive substances that, when combined with synthetic media, enhance cell proliferation and attachment. To maintain its stability, serum must be stored in specialized serum bottles at -5°C to -20°C.

Synthetic Culture Media
Synthetic media are formulated based on the precise requirements of different cell types, containing carbohydrates, amino acids, lipids, inorganic salts, vitamins, trace elements, and cell growth factors. The defined composition allows for better experimental control. However, certain unknown natural components in serum cannot yet be replaced by synthetic substitutes, making it necessary to supplement synthetic media with natural components. The most common practice is adding calf serum to overcome the limitations of synthetic media.
